Detection of Mycolplasma pneumoniae DNA in Sputa of Patients with Atypical Pneumonia Using the Polymerase Chain Reaction

Abstract
The polymerase chain reaction(PCR) method was used to detect Mycoplasma pneumoniae in sputum samples of patients with atypical pneumonia. Two oligonucleotide primers were chosen to amplify a 144 base pairs(bp) frament. In this study 49 samples
from
patients with suspected atypical pneumonia were investigated for PCR. The DNA was released from the bacterial cells by the properties of chaotropic agent guanidinium thiocyanate and was adsorbed by the DNA binding properties of silica particles
without
the steps of organic solvents and ethanol precipitation. A 144-bp DNA fragment was specifically amplified from Mycoplasma pneumoniae genome. The amplified PCR product was detected by ethidium bromide-stained agarose gels. Of 49 sputa tested,
17(34.7%)
were positive for DNA of Mycoplasma pneumoniae. This data suggest sufficiently that Mycoplasma pneumoniae is the important causative agent of atypical pneumonia. Addionally this PCR assay, in our opinion, is rapid and reliable tool for detection
of
Mycoplasma pneumoniae in clinical sputum samples.
